Racura Oncology has entered a research collaboration with Purdue University to investigate how its lead compound (E,E)-bisantrene binds to G-quadruplex DNA structures in the MYC gene promoter to silence MYC transcription in cancer cells. The program, led by G4 structural biology expert Professor Danzhou Yang, will use high-resolution NMR, X-ray and complementary techniques to define the molecular basis of MYC-G4 binding and its impact on regulatory proteins and gene expression.
Early Purdue data indicate that (E,E)-bisantrene directly engages the MYC G-quadruplex to form a stable 2:1 drug–DNA complex, supporting Racura’s strategy of targeting MYC via promoter DNA rather than the historically intractable MYC protein. Over the next two years, the partners aim to generate peer-reviewed, high-impact mechanism-of-action data that can be shared with prospective pharmaceutical partners, potentially strengthening Racura’s scientific credibility and deal-making position in MYC-focused oncology.
