On May 22, 2026, Assembly Biosciences announced that it will expand clinical development of its oral NTCP inhibitor candidate ABI-6250, originally aimed at chronic hepatitis delta virus infection, into primary biliary cholangitis and primary sclerosing cholangitis, extending the program across cholestatic liver diseases. PBC and PSC are chronic autoimmune cholestatic liver diseases marked by impaired bile flow, bile acid buildup and progressive liver damage, for which current treatment options are limited or nonexistent. By blocking the sodium taurocholate co-transporting polypeptide, ABI-6250 aims to modulate bile acid transport and HDV entry, offering a mechanistically distinct oral therapy that could differentiate Assembly Biosciences in the competitive liver disease space if later-stage trials confirm safety and efficacy.
