Alphamab Starts Phase III Trial of Next-Gen HER2 Drug in High-Risk Breast Cancer

Alphamab Oncology ( (HK:9966) ) has issued an update.

Alphamab Oncology announced that the first patient has been dosed in a phase III trial of its bispecific HER2-targeted antibody KN026, co-developed with CSPC subsidiary Shanghai JMT-Bio, in combination with albumin-bound docetaxel (HB1801) and chemotherapy as adjuvant treatment for high-risk HER2-positive breast cancer. The large, randomized multicenter study will compare this regimen with standard trastuzumab plus pertuzumab-based therapy, aiming to improve invasive disease-free survival and other key outcomes for patients with extensive lymph node involvement.

KN026, designed as a next-generation HER2 therapy with dual epitope binding and enhanced tumor-killing mechanisms, is already in multiple late-stage trials and has secured key regulatory designations in China and the U.S. for gastric cancer. HB1801, CSPC’s albumin-bound docetaxel, has shown favorable safety and efficacy profiles in trials, and the new phase III program underscores Alphamab’s push to expand its HER2 franchise and strengthen its competitive position in major solid tumors with significant unmet medical need.
